Trametinib (Mekinist) — MEK Inhibitor for BRAF V600-Mutated Cancer

Trametinib inhibits MEK1/2 in the MAPK pathway, effective in melanoma and lung cancer with BRAF V600 mutations. Usually combined with Dabrafenib.
Targeting the MAPK/MEK Pathway
Trametinib (Mekinist) is a selective MEK1/2 inhibitor acting on the MAPK/ERK signalling pathway, which is frequently over-activated in cancers harbouring BRAF V600E/K mutations. Combining Trametinib + Dabrafenib (BRAF inhibitor) creates dual blockade, overcoming the resistance mechanisms seen with single BRAF inhibitor therapy.
Indications
Advanced melanoma with BRAF V600 mutation (Dabrafenib + Trametinib combination is standard of care). BRAF V600E non-small cell lung cancer. Anaplastic thyroid cancer BRAF V600E (combined with Dabrafenib).
In China
BRAF V600 testing is mandatory in melanoma diagnostic workflows in China. The Dabrafenib + Trametinib protocol is applied at HYGEIA and GoBroad. Patients require BRAF mutation testing before this protocol is considered.
