Osimertinib (Tagrisso) — 3rd-Generation TKI for EGFR-Mutated Lung Cancer

Osimertinib is a 3rd-generation tyrosine kinase inhibitor (TKI) targeting EGFR mutations, including the resistance mutation T790M. Widely used in lung cancer treatment in China.

Mechanism and Advantage

Osimertinib (Tagrisso) is a 3rd-generation TKI targeting EGFR mutations — including ex19del, L858R and especially T790M (the resistance mutation against 1st/2nd-generation TKIs). The drug also crosses the blood-brain barrier, providing efficacy against brain metastases — a major challenge in EGFR+ lung cancer.

Indications and Practice

Used in EGFR+ non-small cell lung cancer (NSCLC) in both early stages (adjuvant post-surgery) and advanced/metastatic disease. In China, EGFR mutation testing is a mandatory step in standard lung cancer treatment protocols.

Lung cancer is prevalent in China and EGFR mutation rates are higher than in Western populations.
