Larotrectinib is an NTRK gene fusion inhibitor effective regardless of tumour tissue type. The tumor-agnostic model opens a new era of genomics-based treatment.
## Tumor-Agnostic Model
Larotrectinib (Vitrakvi) is a TRK (Tropomyosin receptor kinase) inhibitor approved under a tumor-agnostic model — meaning its efficacy is based on the presence of NTRK gene fusions (NTRK1/2/3) rather than the cancer tissue type. A patient with salivary gland cancer, lung cancer or thyroid cancer can all benefit if they carry an NTRK fusion.
## Clinical Efficacy
Overall response rate (ORR) exceeding 75% in pooled clinical trials; durable responses even in heavily pre-treated patients. This is a paradigm shift: no need to identify cancer type — just the genetic mutation.
## In China
NTRK fusion testing is performed at major centres like GoBroad. Patients with NTRK-positive results from NGS testing in Vietnam can continue Larotrectinib or 2nd-generation TRK inhibitor treatment in China.
