Ipilimumab inhibits CTLA-4, amplifying the anti-cancer immune response. Commonly combined with Nivolumab for optimal outcomes.
CTLA-4 Inhibition Mechanism
Ipilimumab is a monoclonal antibody targeting CTLA-4 — a regulatory "brake" receptor on T cells. By blocking CTLA-4, Ipilimumab enhances and prolongs anti-tumour T cell activity. It was the first approved immune checkpoint inhibitor for melanoma.
Combination with Nivolumab
The Ipilimumab + Nivolumab combination (doublet immunotherapy) is now standard in advanced melanoma, high PD-L1 lung cancer, renal cell carcinoma and hepatocellular carcinoma. Response rates exceed monotherapy but immune-related adverse events also increase — close monitoring is required.
